Introduction:
Heelllooooooooooo Traveleeeers! Welcome (back) to the third installment in the completely excellent Borderlands franchise. In Borderlands: Pre-Sequel you can expect much of the same explosively ridiculous combat coupled with the equally ridiculous amount of weapons and loot items. The achievements are largely similar to the previous games and should not pose much of a problem for the majority of Vault Achievement Hunters.
As of release there is a glitch relating to Moonstone. Full details on how to do this can be found in This thread, but basically you can acquire an infinite amount of it. Even if the game get's a patch to fix this you will still be able to do it by clearing your cache and playing offline. This will help immensely with unlocking 'Moxxi's Sampler' and 'Lunar Looter' sooner rather than later as you will need 480 to unlock these 2 achievements. You can also purchase character upgrades with Moonstone and you'll need 600 Moonstone to buy them all. Note that you max out at 500 Moonstone, so you'll need to spend some to get more to be able to afford all of the upgrades.
Doing this glitch may help you shave quite a few hours off your 100% completion time as you will not need to keep looting Moonstone.
Be aware that there are two missable achievements, " Who You Gonna Call? " and "Guardian Guardian." For "Who You Gonna Call?" you will need to complete a particular s ide mission in four player co-op. See it's description for full details. For "Guardian Guardian" you will need to make a choice during a side mission to get the achievement. Choosing wrong will not unlock it. See it's description for full details.
Regarding the potentially missable achievement "Vault Hunter Superior," it should not be missable but some people are reporting that they somehow managed to finish the final mission without ever encountering it. Killing the Opha Superior is one of the final mission requirements, so you should come across it and kill it, but if you have somehow missed it, DO NOT enter the Vault without first having found and killed it to be sure you haven't missed it.
Regarding the glitched achievement, "Moon Mission Meister," it seems this achievement may not unlock if you complete some of the side missions while playing co-op. You can find details in This post. To have the best chance to avoid the glitch you should do all of the missions in your own game as host and turn them all in yourself.
Step 1 - Normal Playthrough:
Well, as you would imagine, begin the game. As stated in the above roadmap, you do not choose a difficulty but the first playthrough is technically classed as the 'Normal' playthrough. Enemies aren't that difficult and will not usually exceed the mid-late 20's in levels. You yourself will likely be around that level as you complete the game for the first time. During this playthrough you should concentrate on as many of the miscellaneous achievements as possible, although you can always return to this playthrough to complete anything that you may have missed.
Be sure to complete all of the side missions for "Moon Mission Meister" as your progress will not carry over into your True Vault Hunter Mode Playthrough and you will have to do them all again. They are much easier on the Normal playthrough. You will need to discover areas that are only accessible during certain side missions too for the various "Discover all Locations..." achievements.
Step 2: True Vault Hunter Mode Playthrough
You unlock 'True Vault Hunter Mode' after completing the game for the first time. This mode is essentially New Game + and the enemies will be at a higher level all the way up to level 50, except the Invincible bosses who will be 2 levels above at level 52. You will need to play through this second playthrough in order to reach level 50 yourself as you will not be able to do so in 'Normal' mode. It is theoretically possible, but it would take you a monumental amount of time to do so. You will also need to play this mode to defeat the 'Invincible Empyrean Sentinel' for the "The Bigger They Are" achievement as you will have to be level 50 to even stand a chance against it.
In this mode your character along with all of your weapons and equipment will carry over but the campaign progress will be reset as you are essentially starting the game again with your high level character.
Step 3: Alternative Characters
You are going to have to play the game as all 4 characters as each one has an achievement tied to them and their action skill. You can do this first and begin the game with each character and play until you unlock their specific achievement, then switch to the next character if you so choose. Doing it this way may give you a better idea of how each character plays and you can then decide on which one you will use for your full playthroughs.
Alternatively you can do this after you have done everything else, quickly getting them unlocked especially if you have little desire to use any of the other characters.
Step 4: DLC - Holodome Onslaught
Welcome to the Holodome! This wave survival mode will have you battling against the biggest and finest enemies that the Moon of Pandora has to offer. There are only three achievements and they can be done in a single sitting. one of the three achievements can prove to be a little tricky but nothing too difficult, especially for the Borderlands veterans among you.
To begin the DLC, after downloading it, you will be able to fast travel to a location called 'The Holodome.'
Step 5 DLC - Claptastic Voyage:
Welcome to the magical world of Claptrap's mind. In Claptastic Voyage you will be sent into the mind of Claptrap tasked with retrieving valuable code for Handsome Jack.
You can expect more of the same Borderlands humour and gameplay, along with a familiar looking set of achievements, including the dreaded wave defence achievements. Not to worry though as they are mostly very easy and straight-forward and can easily be done in a single sitting.
Be aware of the missable achievement "Wheely Fast" and see it's description below for details on how to avoid having to play through the DLC again to obtain it.
To start the DLC go to a fast travel station and travel to a location called 'Deck 13 1/2.' Note that you should be around level 30-35 to play this as the enemies begin at around level 31.

Conclusion:
So, if you played the previous two, you're looking at a relatively similar list of achievements, which are certainly manageable for any lone ranger to solo, but again will be more fun and a lot easier if you decide to go through the game with friends.
